What is SmartSense on the Cannondale Synapse?

SmartSense is Cannondale's integrated system on some Synapse builds, combining front and rear lights with a rear-facing radar that warns of approaching cars, powered by a central battery. It is a genuine safety feature the Roubaix does not match.

Is the Synapse or Roubaix more comfortable?

Both are very comfortable. The Roubaix's FutureShock smooths bumps at the bars, while the Synapse relies on frame design and flex. The Roubaix feels livelier up front; the Synapse feels more planted.

Which should I buy used?

On the Roubaix, check the FutureShock for lost damping. On a SmartSense Synapse, check the lights, radar and battery all work, as that system is costly to fix. Otherwise both are sound used endurance buys.
